Project 1: Understanding the Influence of Glycosylation on Microglial Cells and Alzheimer's Disease

Alzheimer's Disease (AD) poses a significant global health challenge. In this project, we are delving into the intricate world of glycosylation and its impact on microglial cells, which play a pivotal role in neuroinflammation and AD progression. By deciphering the glycosylation patterns that modulate the structure and function of microglial cells, we aim to uncover novel targets for the development of innovative drugs to tackle this debilitating disorder.

Key Objectives : 

- Investigate the glycosylation patterns of microglial cells in AD.
- Uncover how glycosylation influences microglial cell structure and function.
- Identify potential drug targets for AD therapy based on glycosylation insights.

Project 2: Harnessing Glycoengineering for Antibody-Drug Conjugate Development

Antibody-Drug Conjugates (ADCs) have revolutionized targeted cancer therapy. In this project, we're taking ADCs to the next level by targeting sialic acids at the monoclonal antibody's (mAb) Fc region. By introducing terminal sialylation through glycoengineering, we're aiming to create a platform for precise drug conjugation. This innovative approach addresses the challenge of Fc sialylation in recombinant mAbs and paves the way for enhanced ADC efficacy.

Key Objectives:

- Develop glycoengineered mAbs with terminal sialylation for optimal drug conjugation.
- Investigate the site-specific drug conjugation potential at Fc region sialic acids.
- Elevate the effectiveness of ADCs by harnessing glycoengineering techniques.

Project 3: Biopharmaceutical Processing and Characterization

1. Structure-Function Evaluation of Deamidation in Monoclonal Antibodies (mAbs)

  • In Collaboration with Sciex

Monoclonal antibodies (mAbs) are indispensable therapeutic agents, but post-translational modifications like deamidation can impact their efficacy and stability. In collaboration with Sciex, we employ cutting-edge mass spectrometry techniques to elucidate the structural consequences of deamidation in mAbs, ultimately enhancing our ability to design and optimize these life-saving biotherapeutics.

2. Modeling of Chinese Hamster Ovary (CHO) Glycosylation

  • In Collaboration with SymphonyTech Biologics

Chinese Hamster Ovary (CHO) cells are commonly used in biopharmaceutical production. Our collaboration with SymphonyTech Biologics focuses on developing advanced models for CHO cell glycosylation. Mass spectrometry plays a pivotal role in characterizing and predicting glycan profiles, offering valuable insights into bioprocessing and glycoengineering strategies.

We are looking for highly motivated post-doctoral fellows, Ph.D. students, and Project fellows to join our group and work on exciting projects to understand the role of glycans in various diseases. We also develop analytical methods for the in-depth characterization of biopharmaceuticals.  Prior experience in protein expression, cloning, purification, and proteomics/ glycoproteomics is desirable but not essential.
